鱼鳞明胶酶解制备超氧阴离子清除活性肽的研究 被引量:9

摘要 研究了Alcalase2.4L水解鱼鳞明胶的工艺条件及产物的超氧阴离子清除活性。以水解度为指标,在单因素实验的基础上,通过响应面实验优化得到最佳酶解条件为温度54.5℃,pH7.7,底物浓度1.8%(W/V),加酶量1%(W/W),在此条件下水解2h的水解度为13.02%。对酶解产物进行体外超氧阴离子清除活性分析,结果表明,不同水解度的酶解产物表现出不同的抗氧化活性。在优化条件下水解4h,水解产物的超氧阴离子清除活性最高,IC50为0.36mg/mL,此时水解度为14.11%。 The hydrolyzation of fish scales gelatin with protease Alcalase 2.4L and the superoxide radical scavenging activity of hydrolysates were studied.The optimal hydrolytic conditions for degree of hydrlysis (DH) were determined by response surface methodology based on single factor experiments.The results showed that the optimal hydrolytic conditions were temperature 54.5℃, pH 7.7, substrate concentration 1.8% and the ratio of alcalase/substrate 1%.When the fish scales gelatin was hydrolized by Alacalase 2AL for 2h under the optimal conditions,the DH was 13.02%.Whereas,the study on superoxide radical scavenging activity of hydrolysates with different DH under the optimal conditions showed that the hydrolysate had the highest superoxide radical scavenging activity after 4h hydrolysis with DH 14.11%.The 50% inhibitory concentration of the peptides was as high as 0.36mg · mL^-1.
