摘要
目的探讨泽黄颗粒对糖尿病大鼠肾组织nephrin表达的调节。方法用高脂高糖饲料加小剂量STZ诱导糖尿病大鼠模型,随机分为糖尿病模型组(DM组)、泽黄颗粒治疗组(Z组)、格列吡嗪对照组(G组)和正常对照组(N组)。实验第12周末检测BG、Cc(r肌酐清除率)及UAER(尿白蛋白排泄率),观察肾组织病理改变,运用免疫组织化学、Western-Blot、Real-Time PCR法检测肾脏nephrin蛋白及mRNA表达的变化。结果泽黄颗粒可以改善DM大鼠Ccr,降低UAER,改善肾脏病理,减轻足突融合,维持足细胞相关蛋白分子的分布与表达。结论泽黄颗粒通过上调足细胞相关蛋白分子nephrin水平而减轻足细胞损伤,对糖尿病大鼠足细胞损伤有保护作用。
Objective To investigate the regulatory effect of Zehuang Granule (ZG) on renal neprin expression in diabetic rats. Methods Diabetic rat model was induced by fed with high fat and high glucose diet and small-dosage streptozotocin. The rats were randomized into diabetes mellitus (DM)model group(DM group), ZG group(Z group), Glipizide Tablets(G group)and normal control group(N group). We measured the level of blood glucose, serum creati- nine, urine creatinine and urinary albumine excretion rate at the end of the 12 th week, and detected the renal patho- morphological changes under light microscope and electron microscope, lmmunohistochemical staining, Western-Blot and Real-time PCR were adopted to detect the protein and mRNA expression of renal nephrin. Results ZG improved the creatinine clearance rate in diabetic rat s, degraded the urinary albumine excretion rate, relieved the renal patho- morphological changes ,lessened the fusion of foot process, maintained the distribution and expression of podocyte - associated protein molecule. Conclusion It is initially confirmed that ZG can up-regulate the expression of podocyte- associated protein molecule of nephrin, and reduce the injury of podocytes, which is probably one of the protective mechanism of ZG in diabetic rats.
