摘要
目的研究雷公藤多苷对胶原诱导性关节炎(CIA)大鼠血管内皮生长因子(VEGF)基因表达的影响,探讨其在治疗类风湿关节炎中的抗血管生成机制。方法建立大鼠CIA模型,造模成功后灌胃给药,动物分为正常组、模型对照组、沙利度胺组(作为阳性对照)、雷公藤多苷组,采用实时荧光定量PCR法检测大鼠不同组织(肺、心、肝、肾、滑膜)中VEGF的表达,观察雷公藤多苷对VEGF基因表达的影响。结果雷公藤多苷组、沙利度胺组大鼠各组织中VEGF mRNA均低于模型对照组(P<0.05),与沙利度胺组比较无明显差异(P>0.05)。结论雷公藤多苷对CIA大鼠VEGF的表达有抑制作用。
Objective To study the effect of tripterygium glycosides on the expression of vascular endothelial growth factor (VEGF) mRNA in rats with collagen-induced arthritis ( CIA), and investigate the mechanism of tripterygium glycosides on antiangiogenesis in treating rheumatoid arthritis. Methods CIA rat model was established. CIA rats were divided into controlled model group, thalidomide group for positive control, and tripterygium glycosides group. Rats not treated with collagen served as normal group. The effect of tripterygium glycosides on the expression of VEGF mRNA was studied in lung, heart, liver, kidney, synovial membrane respectively from rats by real-time PCR. Results The expression of VEGF mRNA from groups of tripterygium glycosides and thalidomide was lower than that in model-controlled group (P 〈 0.05 ). The difference between tripterygium glycosides and thalidomide was statistically not significant (P 〉 0.05). Conclusions Tripterygium glycosides may inhibit the expression of VEGF mRNA in CIA rats.
