摘要
目的观察槲皮素(Que)对大肠癌细胞增殖、侵袭力的影响,并探讨其可能机制。方法加入不同浓度的Que培养大肠癌细胞SW-480后,用软琼脂克隆培养试验检测SW-480的增殖能力,用Boyden小室模型法检测SW-480侵袭力,用荧光实时定量RT-PCR检测SW-480的滋养层细胞表面抗原2(TROP-2)mRNA,以Western blot法检测SW-480的TROP-2蛋白。结果SW-480加入Que培养后,恶性增殖和穿膜细胞数均明显下降,且呈剂量依赖性(P均<0.01);同时其TROP-2 mRNA和蛋白表达水平均明显下调,且呈时间和剂量依赖性(P均<0.01)。结论Que可明显抑制大肠癌SW-480细胞的增殖、侵袭力,其机制可能与下调TROP-2基因表达有关。
Objective To study the effects and mechanism of quereetin on human eoloreetal cancer cell, Methods Human eolorectal cancer cells SW-480 were treated with different doses of quereetin, and the growth of anchorage independence of SW-480 was studied by colony formation in soft agar, the invasiveness of SW-480 was determined by Boyden chamber, the expression of mRNA and protein of trophoblast cell-surface antigen-2 (TROP-2)gene were detected by real time RT-PCR and Western blot assay, respectively. Results Transmembrane cell and anchorage independence growth of querce- tin group decreased in dose-dependent manners(P 〈0.01 ). The level of mRNA and protein of TROP-2 of SW-480 treated with quercetin reduced in time- and dose-dependent manners(P 〈0.01 ). Conclusions Quercetin can inhibit invasiveness and proliferation of human colorectal carcinoma cells, and decreasing the expression of TROP-2 gene may play an important role in its mechanism.
