摘要
40尾光唇裂腹鱼(Schizothorax lissolabiatus)采自珠江水系可渡河,采用PCR扩增和直接测序技术对其mtDNA D-loop区481bp的序列进行了测定分析。该序列共计发现了10个变异位点,占分析总位点数的2.08%。定义了10种单倍型,单倍型多样度(Hd)为0.733,单倍型间平均遗传距离(P)为0.0051。核苷酸多样性(π)为0.0027,平均核苷酸差异数(K)为1.301。用单倍型间遗传距离构建的NJ树由2个支系组成。该研究显示光唇裂腹鱼可渡河种群mtDNA D-loop区多态性较低,表明光唇裂腹鱼可渡河种群遗传多样性贫乏,有必要采取综合措施开展光唇裂腹鱼可渡河种群的保护。
The sequence polymorphism of mtDNA D-loop in the Schizothorax lissolabiatus population in Kedu River was studied. Fourty individuals of the species were collected form Kedu River in Pearl River system, mt DNA D-loop sequence of 481 bp was analyzed by the methods of PCR and DNA sequencing. A total of 10 varable loci ( 2.08% of total loci) were detected in the sequence of mtDNA D-loop in the population. Ten haplotypes were detected from the 40 individuals. Haplotypic diversity (Hd)was 0.733, and average genetic distance (P)between the haplotypes was 0.005 1. Nucleotide diversity (π) of the 40 individuals was 0.002 7, and average number of nucleotide diffences (K) was 1.301. NJ phylogenetic tree based on the genetic distance between the 10 haplotypes clusted into 2 branches. The results indicated there were poor sequence polymorphism and low genetic diversity in mtDNA D-loop in this population, and therefore the protection for the population is much necessary.
