摘要
目的探讨阿魏酸钠(SF)对糖尿病(DM)大鼠肾脏转化生长因子-β1(TGF-β1)和Ⅳ型胶原表达的影响。方法Wistar大鼠随机分为正常对照组、DM对照组、SF治疗组,制备链脲佐菌素(STZ)诱导的DM大鼠模型,SF治疗组灌胃给予SF110mg/(kg·d),治疗8周。分别测定各组大鼠肾重/体重、肌酐清除率(Ccr)、24h尿蛋白定量,观察肾脏病理改变并用免疫组织化学方法检测肾脏TGF-β1和Ⅳ型胶原的表达。结果与正常对照组相比,DM组大鼠肾重/体重、Ccr及24h尿蛋白均显著升高(P〈0.05);肾小球体积显著增大,系膜区增宽(PAS红染区扩大),且TGF—β1和Ⅳ型胶原表达明显增高(P〈0.05)。SF治疗组上述指标的异常改变均有显著改善(P〈0.05)。结论SF对DM大鼠的肾脏损害具有一定的保护作用,有效的抑制DM大鼠肾脏TGF—β1和Ⅳ型胶原的高表达是其可能的机制。
Objective To investigate the effects of sodium ferulate (SF) on the expression of transforming growth factor-β1 (TGF- β1 ) and collagen Ⅳ in kidneys of diabetic rats. Methods Healthy male Wistar rats were random divided into three groups : Normal control group, strepotozocin induced diabetic rats group and strepotozocin induced diabetic rats treated with SF group. The renal weight/body weight, clearance rate of creatinine (Ccr) and twenty-four hours urinary protein were measured. The pathology changes were observed and immunohistochemical staining were used to detect the expression of TGF-β1 and collagen Ⅳ in renal tissues. Results Compared to normal control group, the levels of Ccr, renal weight/body weight and urinary protein of twenty-four hours were significantly increased in the diabetic control group, and the group were associated with a marked glomerular extracellular matrix accumulation and mesangial expansion ( PAS staining). The expression of TGF-β1 and collagen Ⅳ protein in renal tissues were also up-regulated. SF administration made all changes much better. Conclusion SF administration in diabetic rats can ameliorate the renal damage. Its beneficial effects may contribute to the inhibitory expression of TGF-β1 and collagen Ⅳ in kidney.
