摘要
目的建立高效液相色谱法检测酶促反应液中L-半胱氨酸含量。方法采用异硫氰酸苯酯柱前衍生高效液相色谱法测定。色谱柱:Waters Symmetry C18(5μm,250mm×4.6mm);流动相A:醋酸钠缓冲溶液-乙腈(40:3),流动相B:乙腈-水(4:1);梯度洗脱,流速1.0mL/min;柱温38℃;检测波长254nm。结果线性范围为0.01~0.1mg/mL,r=0.9995(n=6);平均回收率为100.03%,RSD为0.21%(n=6)。结论此法简便易行。
Purpose To establish a HPLC method for the content determination of L-cysteine in enzymatic reaction mixture. Methods L-cysteine was derivatized with phenylisothiocyanate, and the content of L-cysteine in enzymatic reaction mixture was determined by HPLC. The column was Waters Symmetry C18 (5μm,250 mm × 4.6 mm).The mobile phase A was sodium acetate buffer-acetonitrile(40: 3), and the mobile phase B was acetonitrile-water(4:1 ). The flow rate was 1.0 mL/min. Detection wavelength was 254 nm and the column temperature was 38 ℃. Results The linear range was 0.01-0.1 mg/mL, r = 0.999 5( n = 6). The average recovery was 100.03 % ,and RSD was 0.21% (n = 6). Conclusion The method is simple, rapid ,accurate and reliable.
