摘要
目的:观察红花对兔耳增生性瘢痕(HS)结缔组织生长因子(CTGF)表达的影响,探讨红花抑制兔耳HS可能的分子生物学机制.方法:27只新西兰大白兔耳腹侧建立HS模型,每耳2块.建模后45 d开始对HS行注射治疗,1次/wk,连续注射4次.设立阴性对照组、阳性对照组、生理盐水组、低浓度红花(125 g/L)组、高浓度红花(500 g/L)组.注射后第2,4,6周分别切取8只兔耳HS及皮肤,采用免疫组织化学方法(SP)检测每块组织CTGF蛋白的表达(CTGF蛋白面密度及平均吸光度),逆转录聚合酶链反应(RT-PCR)方法检测每块组织CTGF-mRNA的表达.结果:注射后第2,4,6周,两种浓度红花组CTGF蛋白面密度、平均吸光度及CTGF-mRNA的表达均低于阳性对照及生理盐水组,有统计学差异(P<0.05);其中高浓度红花组CTGF蛋白面密度、平均吸光度及CTGF-mRNA的表达为同时段所有HS组中最低(P<0.05).结论:高、低浓度红花均能抑制兔耳HS的CTGF的表达,高浓度红花的作用较低浓度红花强.
AIM:To investigate the effect of carthamus tinctorius on connective tissue growth factor (CTGF) expression in hypertrophic scars (HS) of rabbit's ears. METHODS : Twenty-seven New Zealand rabbits were chosen as experiment animals and the models of HS were established with 2 HS in each ear. On the 45th day after the model establishment, HS were injected with nothing, sodium chloride, carthamus tinctorius (125 g/L) and carthamus tinctorius (500 g/L), respectively once a week for 4 weeks. At the 2nd, 4th and 6th week after injection, HS and skin of 8 rabbits were cut and detected by immunohistochemical method (sp) and reverse transcription polymerase chain reaction (RTPCR). RESULTS: The expressions of CTGF protein and CTGF- mRNA in HS with carthamus tinctorius( 125 g/L) and carthamus tinctorius (500 g/L) were lower than those in positive control group and sodium chloride group at the 2nd, 4th and 6th week after injection( P 〈 0.05 ). The expressions of CTGF protein and CTGF-mRNA in HS with carthamus tinctorius(500 g/L) were the lowest (P 〈 0.05 ). CONCLUSION : Carthamus tinctorius ( 125 g/L) and carthamus tinctorius(500 g/L)inhibit the expression of CTGF in hypertrophic scar of rabbit's ears. The effect of carthamus tinctorius (500 g/L) is stronger than carthamus tinctorius ( 125 g/L ).
