复合酶制备鳕鱼皮梯级胶原肽的初步研究 被引量:17

PREPARATION OF SERIES OF PEPTIDES FROM SKIN OF COD BY COMPOUND ENZYME HYDROLYSIS

摘要 利用酶工程技术,以鳕鱼皮作为研究对象,研究开发鳕鱼皮梯级胶原肽制品。选取胰蛋白酶和碱性蛋白酶作为试验用酶,通过正交试验方法确定最适水解条件,胰蛋白酶:加酶量2.5%、温度45℃、pH6.5、时间4h;碱性蛋白酶:加酶量4%、温度50℃、pH8.0、时间3h。复合酶解的最适条件为:胰蛋白酶和碱性蛋白酶混合水解,加酶量分别为2.5%和4%、温度50℃、pH8.0、时间4h,蛋白质水解度26.1%。分离得到两多肽组分UF-1和UF-2,分子量(Mr)范围分别为:UF-1:2000u<Mr<6000u;UF-2:Mr<2000u,其中UF-2的含量最高为83.32%。用高效液相色谱分析水解产物相对分子量分布,确定复合酶解产物为蛋白胶原肽和游离氨基酸,其相对分子质量集中分布在1350u和145u左右。 The research studied the preparation of series of peptide from skin of Walleye Pollock by compound enzyme hydrolysis. The appropriate protease and condition of hydrolysis was selected through the orthogonal tests. The optimum parameters of trypsin were: enzyme-to-substrat ratio of 2.5 %, the temperature of 45℃, pH 6.5 and 4 h; The optimum parameters of Properase E were: enzyme-to-substrat ratio of 4 %, the temperature of 50℃, pH 8.0 and 3 h. The optimum parameters of the compound enzyme hydrolysis were: enzyme-to- substrat ratio of 2.5 % and 4 % respectively, the temperature of 50 ℃, pH 8.0 and 4 h, and the hydrolysis was 26.1%. Hydrolysates were separated into two parts by ultra-filtration technology, UF-1 and UF-2. The molecular weights of UF-1 and UF-2 were 2 000 u〈Mr〈6 000 u and Mr〈2 000 u respectively. The yield of UF- 2 was 83.32 %. HPSEC was used to determine the molecular weight distribution of hydrolysates, the results indicated that the peptide molecular weights hydrolyzed by mixed proteases were about 1 350 u and 145u, respectively.
