摘要
目的建立奥沙利铂脂质体包封率的测定方法。方法用阳离子交换树脂分离游离药物和被包封药物,采用高效液相色谱法测定奥沙利铂含量,计算包封率。结果阳离子树脂对溶液中游离奥沙利铂的吸附率大于93%,对载药后的脂质体无吸附;HPLC含量测定方法的线性范围为1~100mg·L-1(R2=0.9999),精密度RSD<2%。结论阳离子树脂-HPLC法简便快捷,成本较低,且无空白干扰,方法学稳定,适用于奥沙利铂脂质体包封率的测定。
Objective To establish a method for determining the entrapement of oxaliptation liposomes; Method The free oxaliplatin and its liposomes were separated by using cationic exchange resin,and the concentration of oxaliplatin was determined by HPLC; Result Cationic exchange resin's absorption rate of oxaliplation in solutions was more than 93 %, and cationic exchange resin had no absorption of the liposome loading oxaliplation. The linearity range of assaying was 1-100 mg · L^-1 , the precision evaluated by RSD was Less than 2 % ; Conclusion The established method for determination the entrapement efficiency of oxaliplatin liposomes is simple, rapid and cheap.
