摘要
目的:优选印楝叶总黄酮的提取工艺条件,并建立总黄酮的含量测定方法。方法:通过正交试验,考察溶剂浓度、回流时间和回流次数对提取工艺的影响。采用分光光度法,以芦丁为对照品,在波长500nm处测定药材总黄酮含量。结果:最佳提取条件为80%的乙醇,回流提取3次,每次1h。总黄酮在0.020~0.080mg/mL范围内线性关系良好(R=0.9992),平均加样回收率为96.67%,RSD为0.96%。结论:优选出的最佳工艺条件经验证其结果稳定;总黄酮含量测定方法简便、快速、稳定,可用于测定印楝叶中总黄酮的含量。
Objective: To optimize extraction technique of total flavonoids and establish a method for measuring the contents of total flavonoids in the leaves of Azadirachta indica. Methods: The optimum process was investigated by L9 (34) orthogonal design. The contents of total flavonoids in the leaves of Azadirachta indica were measured using speetrophotometry with a detection wavelength of 500nm, borrowing the Rutin as a contrast. Results: The optimum technical condition: 80% ethanol, refluxing and extracting for 3 times, one hour each time. The linear range was between 0. 020 ~0. 080mg/mL (R =0. 9992) and the average recovery was 96. 67% with corresponding RSD of 0. 96%. Conclusion: The optimum extraction technology of total flavonoids is stable. The method for determination is simple, rapid and is suitable for determination of total flavonoids in the leaves of Azadirachta indicant.
