摘要
一般认为分蘖由QTL控制,对植物侧生组织的研究表明,水稻分蘖的形成可能受激素的控制。以寡分蘖突变体G069与广亲和材料"02428"构建的1对近等基因系02428-ftl为对象,采用RNA差异显示技术分析了此对近等基因系在分蘖前期的基因差异表达。经过片段回收,反式Northern杂交(Re-verse northern blotting)和测序分析,分离得到一个在"02428"中强势表达的基因片段。生物信息学分析表明,该片段与水稻的cDNA和mRNA以及基因组序列高度相似,但没有同源的氨基酸序列,说明该片段处于非编码区;电子延伸序列与多个水稻多肽序列高度同源,含有一个明显的蛋白激酶作用域,但未发现与已报道的基因同源。推测为一未知的调控蛋白。
Tiller was generally considered being controlled by QTLs. Some studies on lateral meristem revealed that the tiller of vile maybe regulated by phytohormone. We conducted the study on a NIL (near isogenic line),02428-ftl,with differential display PCR (DD-PCR)teehnology, and isolated a segment refer to an unknown gene which expressed with higher level in wild type (02428) than in the mutant (ftl). Bioinformatic analysis showed that there was high identity between the segment and many other rice sequences including eDNA, RNA and genome sequences in rice database. The corresponding protein was putative as a regulatory protein for its obvious kinase domain.
