福州地区汉族老年男性维生素D受体基因多态性与骨密度的关系 被引量:2

The relationship between polymorphisms of vitamin D receptor gene and bone mineral density in Fuzhou elderly male

摘要 目的观察福州地区汉族老年男性维生素D受体(vitaminDreceptor,VDR)基因BsmI,ApaI及TaqI多态性分布及与骨密度的关系。方法选择福州地区60岁以上汉族男性150例,检测其正位第2—4腰椎、左侧股骨颈、大转子和Ward’s区骨密度,应用聚合酶链式反应一限制性片段长度多态性(PCR—RFLP)检测维生素D受体基因BsmI,ApaI及TaqI多态性。结果受试对象中,VDRBsmI基因型分别为BB型5例(占3.3%)、Bb型11例(占7.3%),bb型134例(占89.4%),b等位基因频率为93.0%、B等位基因频率为7.0%;ApaI基因型分别为AA型12例(占8.0%)、Aa型64例(占42.7%),aa型74例(占49.3%),A等位基因频率为29.3%、a等位基因频率为70.7%;TaqI基因型分别为1Tr型136例(占90.7%)、Tt型14例(占9.3%),无tt型,T等位基因频率为95.3%、t等位基因频率为4.7%。分析基因型与骨密度的关系显示:三个多态性位点各基因型人群在腰椎、股骨颈、大转子、Ward’s区4个部位骨密度差异无统计学意义(P〉0.05)。结论维生素D受体基因BsmI,ApaI及TaqI多态性与骨密度间没有关联,尚不能作为预测福州地区老年男性发生骨质疏松危险性的遗传标志。 Objective To investigate the prevalence of vitamin D receptor (VDR) gene polymorphism in Fuzhou elderly male, and to study the association between Bsml, Apal, Taql polymorphism of VDR gene and bone mineral density (BMD). Methods BMD of lumbar spine, femoral neck, troch and Ward's triangle were measured by dual-energy X-ray absorptionmetry. The genotypes of VDR gene of 150 male was detected by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). Results The distribution of VDR Bsml genotypes in 150 male was BB genotype 5(3.3%), Bb 11(7.3%), bb 134(89.4%) and the b allelic gene frequencies reached 93.0% while B was 7.0%. The distribution of VDR Apal genotype was AA genotype 12(8.0% ), Aa 74(42.7% ), aa 6,4(49.3% ), respectively. The A allelic gene frequencies reached 29.3% while a was 70.7%. There were only two genotypes found among the people with Tr genotypes 136 (90.7%) and Tt 14 (9.3%). The T and t frequencies were 95.3% and 4.7% respectively. There was no significant difference in BMD of lumbar spine, femoral neck, troch and Ward's triangle among genotypes of each polymorphism site. Conclusion There was no significant relationship between gene polymorphism of VDR and BMD in Fuzhou elderly men, and it is not regarded as a genetic marker in predicting the risk of developing osteoporosis in Fuzhou elderly male.
