摘要
目的:探讨不同浓度吡咯烷二硫氨基甲酸(PDTC)对高糖下大鼠成纤维细胞(NRK)中单核细胞趋化蛋白-1(MCP-1)表达的影响。方法:体外培养大鼠NRK细胞;葡萄糖孵育下分6组,正常对照组、高糖1组、高糖2组含葡萄糖分别为5.6、15和30mmol/L,干预组在葡萄糖30mmo/L基础上分别加入PDTC5、10和20μmol/L;RT-PCR检测24、48h后各组MCP-1mRNA表达水平,免疫细胞化学(ICC)法检测24、48h各组的MCP-1蛋白表达情况。结果:与正常对照组相比,高糖组MCP-1mRNA和蛋白表达水平显著升高(P<0.05),且呈剂量时间依赖性;而不同浓度PDTC干预后,MCP-1mRNA和蛋白表达水平显著下降(P<0.05),且与PDTC呈剂量时间依赖性。结论:PDTC下调NRK细胞中MCP-1mRNA和蛋白的表达。
Objective To explore the effect of different concentrations of PDTC on the expression of monocyte chemotactic protein-1 (MCP-1) in rat kidney fibroblasts (NRK) under high glucose circumstance. Methods NRK were cultured in vitro, and then were assigned to receive 5.6 mmol/L glucose (group 1), 15 mmol/L glucose (group 2), 30 mmol/L glucose (group 3), 30 mmol/L glucose + 5 μmol/L PDTC (group 4), 30 mmol/L glucose + 10 μmol/L PDTC (group 5), or 30 mmol/L glucose + 20 μmol/L PDTC (group 6). The expression of MCP-1 mRNA was measured by RT-PCR and MCP-1 was measured by ICC after 24 h and 48 h, respectively. Results As compared with group 1, MCP-1 mRNA and MCP-1 were remarkably increased in a time-dose dependent manner in group 2 and group 3 (P 〈 0.05). MCP-1 mRNA and MCP-1 were remarkably decreased in a time-dose dependent manner in group 4, group 5, and group 6, as compared with those in group 3(P 〈 0.05). Conclusion PTDC can downregulate the expression of MCP-1 mRNA and MCP-1 in NRK.
