新生隐球菌共孵育后血管内皮细胞的蛋白质组学研究 被引量:1

摘要 目的研究新生隐球菌共孵育后血管内皮细胞与正常细胞的差异蛋白质谱,推测蛋白质表达改变在孵育过程中的作用。方法利用二维凝胶电泳获得新生隐球菌孵育后血管内皮细胞与正常细胞的差异表达蛋白点,对部分差异蛋白点进行质谱鉴定分析,并以实时荧光定量PCR对其mRNA表达进行定量比较。结果Peroxiredoxin Ⅰ及Calpactin Ⅰ light chain等13个蛋白的表达水平发生明显改变,Peroxiredoxin Ⅰ及Calpactin Ⅰlight chain的mRNA表达明显改变,其mRNA含量的改变趋势与相应的蛋白质的变化趋势相同。结论Peroxiredoxin Ⅰ及Calpactin Ⅰ light chain等蛋白表达量的改变可能与新生隐球菌侵袭血管内皮细胞屏障有关。 Objective To study the differences of protein profile between HUVEC incubated with Cryptococcus neoformans and normal HUVEC,and to speculate its role during incubation. Methods The expression proteins between HUVEC incubated with Cryptococcus neoformans and normal HUVEC were obtained by 2-D electrophoresis and the differences were identified by MALDI- TOF MS technology, mRNA was quantitated by Real-time q-PCR. Results The expression level of 13 kinds of protein changed evi- dently,especially Peroxiredoxin I and Calpactin I light chain,with corresponding changes of mRNA quantity. Conclusion Expression changes of Peroxiredoxin I and Calpactin I light chain may have relationship with the invasion of Cryptococcus neoformans into the barrier of HUVEC.
