摘要
目的通过比较两种从石蜡包埋组织中提取基因组DNA的方法,确定一种高效、经济而简便的方法指导实际工作。方法按照两种方法(脱蜡法与非脱蜡法)提取DNA,比较所提DNA的质量,PCR扩增β-Globin基因作为基因分析的对照扩增,并对其中非脱蜡法进行改良。结果两种方法所提取的DNA均能达到相关分子学实验的要求,PCR扩增带清晰,结果较为相似。结论非脱蜡法也能提取出适用于PCR技术要求的DNA,方法简便、经济。
Objective To seek an effective,economic and simple DNA Extraction methods from Unbufered Formalin-Fixed and Paraffin-Embedded tissue, two methods were compared in our study. Methods Two DNA Extraction methods （phenol/chloroform purification and Goelz＇method） to obtained DNA,and compared the quailty of DNA by analyzed with electrophoresis and PCR amplification. Modification was made to the Goelz＇method. Results All of the two methods could give high quality DNA, the results of PCR were resemblance, but they were diffremt in terms of time-and lab input. Conclusion The Goelz＇method was simple and reliable when it is considered applied in PCR-bases studies.
