人成纤维样滑膜细胞冻存时间对其活性的影响

摘要 目的探讨适合人成纤维样滑膜细胞(FLS)低温冷藏保存的最佳冻存时间。方法将FLS经-80℃冰箱冷冻保存。在冷冻后2、3、4、5wk分别复苏,培养。观察FLS的生长情况和细胞存活率及24h贴壁率。结果冻存5wk的FLS细胞存活率(66±1.3)%,较冻存2wk(90±2.0)%有明显下降;冻存5wk的FLS24h贴壁率(61.3±2)%,与冻存2wk(85.9±3)%比较有明显下降;未冻存和冻存2wk复苏后FLS的增殖能力无明显差异。结论FLS可以进行低温冷冻,冻存时间4wk为宜。 Aim To determine appropriate length of cryopreservation time for fibroblast-like synoviocytes. Methods FLS were cryopreserved in - 80℃ refrigerator, resuscitated and cultured after being frozed for 2,3,4 and 5 weeks. The cell viability and 24 hours adherence rate were evaluated. Results There was significant difference in the rate of cell livability of FLS cryopreserved for 5 weeks compared with that for 2 weeks. Significant difference was also observed in the 24 hours adherence rate of FLS cryopreserved for 5 weeks compared with that for 2 weeks. There was no difference in the proliferation of FLS routinely maintained compared with those cryopreserved for 2 weeks. Conclusions FLS can be cryopreserved and the time limit of cryopreservation should be 4 weeks.
