琐琐葡萄提取物多糖、黄酮对肝癌细胞HepG2的增殖抑制作用 被引量:6

Study on the inhibiting capacity of flavonoid and polysaccharide extracted from SuoSuo Grape to HepG2 Cells' reproductive activity

摘要 目的:研究琐琐葡萄提取物多糖(VTP)、黄酮(VTF)在体外对肝癌细胞HepG2增殖的抑制作用。方法:用不同VTP、VTF剂量组对体外培养的HepG2细胞分别干预24、48、72h后,用MTT法来观察其对HepG2细胞增殖的抑制作用。结果:(1)琐琐葡萄提取物VTP、VTF对HepG2细胞增殖在24h未表现出抑制作用;(2)与阴性对照组相比,除72hVTP剂量组(12500mg/L)外,48h和72h其余剂量组均表现出对HepG2细胞株的抑制作用,其作用差异有统计学意义(P<0.05);VTP和VTF所有剂量组48h均显出统计学意义(P<0.001);(3)重复测量设计资料的方差分析证明了琐琐葡萄提取物VTP、VTF对肝癌细胞株HepG2细胞的增殖有一定的抑制作用,测定时点与药物剂量分组存在交互作用;综合时间抑制曲线,琐琐葡萄提取物VTP和VTF在48h对肝癌细胞HepG2的抑制效果最佳。结论:通过体外对肝癌细胞株HepG2增殖的抑制初步证实,琐琐葡萄提取物多糖和黄酮具有一定抑制肝癌细胞增殖的作用。 Objective: To observe the inhibiting effect on flavonoid (VTF) and polysaccharide (VTP) extracted from SuoSuo Grape to HepG2 Cells' reproductive activity by MTT method in vitro. Methods: HepG2 cells were cultured in vitro and interfered by different concentrations of VTF and VTP for 24, 48, 72 hours respectively. The inhibition of VTF and VTP extracted from SuoSuo Grape to HepG2 cells were detected by MTT Method. Results: (1) The HepG2 cells kept proliferating in 24 hours in all groups. (2) The VTF and VTP extracted from SuoSuo Grape show the inhibition to HepG2 cells compared with the negative control group at 48h and 72 h (P〈0.05), except 72 h VTP (12 500 mg/L). (3) Repeated analysis proved that VTP and VTF extracted from SuoSuo Grape have an inhibiting effect on the proliferation of HepG2 cells. At the same time an interaction effect between time and dose can be observed..Conclusion: The results proved the inhibition of VTP and VTF. VTF and VTP extracted from SuoSuo Grape show the best inhibiting effect on the proliferation of HepG2 cells at 48 h.
