克氏针茅等位酶分析及其与气候因子的相关关系 被引量:5

摘要 【目的】了解克氏针茅(Stipa krylovii)种群的遗传结构,分析其种群遗传变异的成因和机制。【方法】采用居群遗传学方法及等位酶技术,对内蒙古中东部草原5个克氏针茅种群的遗传多样性进行研究,并分析其与气候因子的相关性。【结果】从18个等位酶系统中筛选得到多酚氧化酶(PPO)、天冬氨酸转氨酶(AAT),酯酶(EST)和过氧化物酶(POD)等4个酶谱清晰的等位酶系统,分析得到11个位点27个等位基因,多态位点比例达100%。5个克氏针茅种群之间的遗传分化系数为0.083 6;种群的遗传距离与实际地理距离之间相关性不显著。克氏针茅等位酶位点部分遗传多样性指标和部分等位基因频率与气候因子的相关性达显著或极显著水平。【结论】克氏针茅种群内存在丰富的遗传多样性,气候因子的选择作用在克氏针茅遗传分化中起重要作用。 【Objective】The research is to understand the genetic structure of Stipa krylovii and to clarify the reason causing the population genetic division.【Method】Populatoin genetics and allozyme were used to research the genetic diversity of Stipa krylovii among five populations in the middle east of Inner Mongolia steppe and analyze the relationship between genetic diversity and the climatic factors.【Result】Among eighteen allozyme systems detected,four allozyme systems with clear bands were analyzed.They were PPO,POD,AAT,EST respectively.The four allozyme systems revealed 11 loci and 27 allele genes.The percentage of polymorphism was 100%.The index of gene division Fst among five S.krylovii populations was 0.083 6;there was no a significant correlation between Nei′s unbiased genetic distance and actual geographic distance of populations.Some genetic diversity indexes of S.krylovii allozyme loci have significant or highly significant correlations to climatic factors,so did some allele gene frequencies.【Conclusion】There were rich genetic diversities in S.krylovii poulations and the climatic factors played an important role for the genetic division among S.krylovii populations.
