摘要
研究软骨多糖对荷瘤小鼠的作用,并探讨其对免疫功能的影响。采用小鼠肉瘤S180细胞建立动物腹水瘤模型,然后随机将小鼠分为空白组,模型组和软骨多糖给药组,连续腹腔注射生理盐水或软骨多糖,用单向琼脂扩散免疫实验测定小鼠抗体,用酶联免疫吸附剂法(ABC-ELISA)测定小鼠外周血细胞因子IL-2、IL-6和TNF-α。在软骨多糖的作用下,荷瘤小鼠分泌的IgG抗体明显增加,IL-2的含量由(160.1±21.24)pg·mL-1降至(120.84±12.05)pg·mL-1,IL-6的含量由(68.32±10.43)pg·mL-1升至(99.34±30.93)pg·mL-1,TNF-α的含量由从(0.08±0.019)pg·mL-1提高到了(0.16±0.006)pg·mL-1,从而增强了小鼠的免疫力。由此得出结论,软骨多糖能通过增强S180荷瘤小鼠的体液免疫功能而抑制肿瘤的生长。
Effect of cartilage polysaccharide on the humoral immunity of S180 tumor-bearing mice was studied in this paper. Mouse ascites tumor model was set up using mice sarcoma180 cells and the tested mice were randomly divided into 3 groups (control, model and treated group). Saline or cartilage polysaccharide was injected into the mice by i.p. method. Humoral immunity was detected by single immunodiffusion test and cytokine (IL-2, IL-6 and TNF-α) of peripheral blood was tested by ABC-ELISA method. Single immunodiffusion test showed that IgG antibody was significantly promoted in tumor-beating mice under the effect of cartilage polysaccharide. The content of the IL-2 decreased from (160.1±21.24)Pg·mL-1 to (120.84±12.05) Pg·mL-1, while the content of IL-6 and TNF-α increased from (68.32±10.43, 0.08±0.019)Pg·mL-1 to (99.34±30.93, 0.16±0.006)Pg·mL-1. It was concluded that the cartilage polysaccharide can improve humoral immunologic function of S180 tumor beating mice and inhibit the tumor growth.
