摘要
目的:研究氧化苦参碱在大鼠福尔马林致痛模型中的镇痛作用以及对脊髓c-fos表达的影响,为了进一步阐明其镇痛作用机理,用放射性配受体法研究氧化苦参碱和阿片受体的亲和力。方法:大鼠腹腔给予马钱子碱,测定福尔马林足底注射后疼痛反应指标并用免疫组化方法测定马钱子碱对脊髓c-fos表达的影响。以大鼠大脑为膜蛋白,用3H-二氢吗啡([3H]DHM)为放射性配基,测定氧化苦参碱和阿片受体的亲和力。结果:氧化苦参碱抑制福尔马林引起的I相和Ⅱ相反应(P<0.01),氧化苦参碱组Fos样免疫阳性神经元数明显少于生理盐水组(P<0.01)。放射性配受体实验发现,氧化苦参碱置换放射性标记配基[3H]DHM的IC50大于10-4mol/L。结论:氧化苦参碱能抑制福尔马林引起的急性疼痛和继发性炎症反应以及减少疼痛引起的脊髓水平Fos的表达,有明显的镇痛作用。用放射性配受体法研究发现氧化苦参碱的镇痛作用可能和阿片受体无关。
Objective：To observe the antinocieeption of oxymatfine in the formalin test and its effect to c -fos expression in the dorsal horn of spinal cord in rats. Opioid receptor antagonist used in vivo and radioligand binding assay performed in vitro examined the effects of opioid receptor in brain during the antinocieeption of oxymatrine. Methods ： The formalin test was used to assess the antinocieetion of oxymatrine given intraperitoneaily and immunohistoehemistry technique was used to measure the effect of oxymatrine on c-los expression in spinal cord in rats. Radioligand binding assay was used to determine the aflqnity of brueine and opioid receptor in rat brain membranes displacement studies. Results： Oxymatrine inhibited the reaction of the first and second phase in the formalin test（ P 〈 0.01 ） , and the Foslike immu- noreaetive positive neurons in the group oxymatrine were significantly less than the group saline（ P 〈 0.01 ）. The IC50 of oxymatrine producing displacement of [ 3H ] diprenorphine binding to opioid receptor was bigger than 10-4mol/L. Conclusion： Oxymatrine given intraperitoneallv inhibited both neurogenic and inflammatory phases of the overt noeiception caused by intraplantar injection of formalin and the c-los expression in the dorsal horn of spinal cord in rats. The mechanism of central antinoeieeption seemed not related to opioid receptor. Oxymatrine failed to alter the binding sites of [ 3H ] diprenorphine in vitro.
