湿热毒盛证糖尿病坏疽血管组织中差异表达基因的研究 被引量:2

摘要 目的:研究湿热毒盛证糖尿病坏疽血管组织中的差异表达基因。方法:应用功能分类基因芯片筛选湿热毒盛证糖尿病坏疽血管组织中的差异表达基因,用RT-PCR对部分差异表达基因进行验证。结果:在检测的113个基因中,有13个基因的表达水平上调2倍以上,3个基因的表达水平下调2倍以上,RT-PCR检测了其中3个基因,与芯片检测的结果一致。结论:涉及多种基因表达异常的血管组织病变在湿热毒盛证糖尿病坏疽的发生发展过程中占重要地位,分析这些差异基因的表达,有助于全面了解湿热毒盛证糖尿病坏疽发生的确切机制。 Objective:To study the differentially expressed genes in the vascular tissue of diabetic gangrene. Methods:The differentially expressed genes in the vascular tissue of serious diabetic gangrene were screened by the functional classiflc gene chip. Using reverse transcription polymerase chain reaction (RT-PCR) detection to verify the part of the differentially expressed genes. Results:In the detection of 113 genes, 13 of the gene expression level increased more than 2 times,3 of the gene expression level decreased more than 2 times. 3 genes were verified with RT-PCR detection, the results like with chip testing. Conclusion:The abnormal gene expression in a variety of vascular lesions has an important role in the course of development of diabetic gangrene. To analyze these differences in gene expression that contribute to a comprehensive understanding of diabetic gangrene occurred in the exact mechanism.
