摘要
目的探讨金莲花黄酮对人乳腺癌MCF-7细胞株细胞增殖抑制作用及对端粒酶活性的影响。方法不同浓度金莲花黄酮作用于MCF-7细胞24h,以CCK-8法检测细胞增殖抑制作用、流式细胞术检测端粒酶活性。结果9.516、6.344、3.172、1.586、0.793mg/ml5个不同浓度金莲花黄酮作用于MCF-7细胞24h,对细胞增值的抑制率依次为17%、28%、48%、50%、55%。各实验组A490与对照组比较差异均显著(P<0.05,P<0.01),且用药浓度与抑制率呈正相关;端粒酶活性均有不同程度降低、且端粒酶活性与用药浓度成正比。结论金莲花黄酮对MCF-7细胞有明显抑制作用并降低端粒酶活性。
Objective To investigate the inhibition effects and telomerase activity of Trollius flavonoids on human breast cancer cell line MCF-7. Methods MCF-7 cells were treated by Trollius flavonoids of different concentrations for 24 h. The inhibition effect of cell proliferation was detected by CCK-8 method, and the telomerase activity was detected by flow cytometry. Results The inhibition rates of MCF-7 cell proliferation were 17%, 28%, 48%, 50%, 55% after treatment with different concentrations of flavonoids Trollius (9.516, 6.344, 3.172, 1.586, 0.793 mg /ml, respectively) for 24 h, and there was significant difference between experimental and control groups. There was positive correlation between drug concentration and inhibition rate. The telomerase activity was reduced in some degree, and was directly proportional to the concentration of the drug. Conclusions Trollius flavonoids has significant inhibition effect on MCF-7 cells and reduces the telomerase activity.
