摘要
[目的]建立三七总皂甙的液相色谱定量分析方法,研究三七总皂甙的稳定性。[方法]以三七总皂甙为考察指标,采用高效液相色谱法,探讨不同pH值、温度对三七总皂甙稳定性的影响。色谱条件如下:色谱柱为Kromasil C18(4.6 mm×250 mm,5μm);流速为1.00ml/min;检测波长为197 nm;柱温为室温,采用二元梯度洗脱,流动相为乙腈-水。[结果]三七总皂甙液相色谱定量方法具有较强的针对性和准确性。三七总皂甙在pH值>4,低温条件下,比较稳定,而在强酸高温下受影响较大,在pH值为3时,总皂甙含量降低约32.5%,在100℃下,总皂甙含量降低约12.5%。[结论]三七总皂甙在弱酸弱碱,低温条件下,比较稳定,而在强酸高温下受影响大。
[Objective] The study aimed to establish the quantitative analysis method with I-IPLC on total saponin of Panax notoginseng and research the stability of the total saponin. [Method] With the total saponin of P. notoginseng as the investigated index, the HPLC method was used to discuss the effect of different pH value and different temperature on the stability of the total saponins of P. notoginseng. The chromatoraphic conditions were as follows: chromatographic column of Kromasil C18(4.6 mm ×250 mm,5 μm), flow rate at 1.00 ml/min, detection wavelength at 197 nm and colunm temper- ature of room temperature, by using the mobile phase of acetonitrile-water with duality gradient elution. [ Result ] The analytical method with HPLC quantitative measurement on total saponin of P, notoginseng was precise and accurate. The content of total saponin of P. notoginseng was stabile at pH 〉 4 and low temperature, but that was greatly affected under the strong acid and high temperature. The content of P. notogintseng was decreased by nearly 32.5% at pH of 3 and by 12.5% at 100℃. [ Conclusion] The total saponin of P. notoginseng was stable under the weak acid and weak base and low temperature condition, but that was greatly affected under the strong acid and high temperature.
