摘要
[目的]介绍一种适合于富含多糖、多酚植物材料的RNA提取方法。[方法]采用改进的异硫氢酸胍法提取植物材料的RNA,用紫外分光光度计测定其浓度和纯度,并通过RT-PCR进一步检测其质量。[结果]提取总RNA中的28、18、5s条带清晰,比例适中,OD260nm/OD280nm。为1.8—2.0。以提取的总RNA反转录合成的cDNA为模板,能扩增出预期长度的片段,无非特异性产物。[结论]该方法简便易行,成本较低,适合于富舍多糖、多酚植物材料的RNA提取。
[Objective] The study aimed to introduce a kind of method on extracting RNA from plant materials rich in polysaccharide and polyphenol.
[Method] The improved guanidine isothiocyanate method was used to extract RNA from plant, and the purity was used to determine with UV spectrophotometer. And its quality was further determined through RT-PCR. [Result] The 28, 18 and 5 S bands in the RNA extracted had clear bands and proper proportion, with OD260nm/280nm. Taking the synthesized cDNA of the extracted total RNA though reverse transcription as template, gene
sequence with expected could be amplified and non-specific products were not seen. [Conclusion] The method was simple, convenient and low cost. It
was suitable for extracting RNA from plant materials rich in polysaccharide and polyphenol.
