摘要
[目的]为准确鉴定黑米提取物中花青素的成分和含量提供一种有效的测定方法。[方法]用HPLC法测定黑米提取物中花青素的主要成分及其含量。[结果]在黑米提取物的1%盐酸甲醇溶液的紫外-可见吸收光谱的紫外和可见光区各有一个较强的吸收峰,其波长分别为270和520 nm。黑米提取物的4个批次产品在13.551 min附近均有峰检出,它们的保留时间分别为13.722、13.633、13.517和13.518 min,其平均保留时间为13.598 min,与矢车菊素-3-葡萄糖苷标准品的保留时间比较吻合。黑米提取物中矢车菊色素的平均含量为41.8%。3个浓度的矢车菊素-3-葡萄糖苷的平均回收率为104.5%。矢车菊素-3-葡萄糖苷的峰面积的RSD为1.56%(n=5)。[结论]黑米中花青素的主要成分是矢车菊素-3-葡萄糖苷,该测定方法准确可行。
[Objective] The aim was to provide an effective determination method for accurately identifying the component and its content of anthocyanidin in the extracts of black rice.[Method] The main component and its content of anthocyanidin in the extracts of black rice were determined by HPLC method.[Result] In the UV and visible area of UV-vis absorption spectrum of the 1% hydrochloric acid-methanol solution of balck rice extracts,there were 2 stronger abdorption peaks and their wavelengths were 270 and 520 nm resp.There were peaks detected from 4 batches of products of balck rice extracts near 13.551 min,their retention time were 13.722,13.633,13.517 and 13.518 min and their average retention time was 13.598 min,which was relatively consistent with the retention time of standard cyanidin-3-glucoside.The average content of cyanidin in the extracts of black rice was 41.8%.The average recovery of cyanidin-3-glucoside at 3 concn.was 104.5%.The RSD of peak area of cyanidin-3-glucoside was 1.56%(n= 5).[Conclusion] The main component of anthocyanidin in black rice was cyanidin-3-glucoside and this determination method was accurate and feasible.
