摘要
[目的]开展云南野生月季花抗虫物质总黄酮含量的研究,为更好地利用云南野生月季种质资源和月季抗虫新品种的选育打下一定基础。[方法]以云南野生月季花为材料,采用微波辅助萃取(MAE)、分光光度法测定野生月季花样品中的总黄酮含量。[结果]研究确定了微波辅助萃取法提取月季花中黄酮类化合物的最佳条件为:使用60%乙醇,在液固比40∶1条件下,以255 W的功率微波辐射5m in。所得结果相对标准偏差为1.32%(n=6),回收率在97.3%~102.4%。[结论]该方法操作简便,结果可靠,重现性、稳定性好,可广泛用于各种作物中抗虫黄酮类物质含量的研究。
[Objective] The research on the content of the insect-resistant material-flavonoids in wild rose in Yunnan was carried out,which could lay a certain foundation for the better use of wild rose germplasm resource and new rose variety breeding in Yunnan.[Method] The content of the total flavonoids in the samples of wild roses in Yunnan was determined with the method of microwave-assisted extraction(MAE) and spectrophotometry.[Results] The best condition of the method of microwave-assisted extraction of flavonoids was the ethanol,60%,the liquid-solid ratio,40∶1,the microwave power,255 W and the microwave radiation,5 min.The results of the relative standard deviation was 1.32%(n=6),the recovery rate was from 97.3% to 102.4%.[Conclusion] This method was simple,reliable,good reproducibility and stability,which could be used in a wide variety of the research on the content of flavonoid,one kind of insect-resistant,in crops.
