摘要
目的:主要探索T-2毒素对体外培养软骨细胞致损临界值以及硒保护作用。方法:应用单层培养传代软骨细胞,用生化方法检测软骨细胞DNA量和蛋白量,透射电子显微镜观察软骨细胞超微结构。结果:(1)T-2毒素在0.001~0.005mg/L培养液中(以下相同)剂量范围内,随浓度增加,T-2毒素对软骨细胞DNA合成和细胞分裂增殖影响逐渐增大,加硒后该影响有所减弱,但仍不能阻止损伤的发生;(2)T-2毒素在0.0005mg/L时,对软骨细胞的影响甚微;T-2毒素在0.001mg/L和0.005mg/L时,细胞损伤明显,与对照组比较有显著差别(P<0.001)。
In the present investigation,we observed the critical value of the damage of T2 toxin to chondrocytes and the protection of Se.The amounts of DNA and protein in the singlelayer cultured chondrocytes were assessed by the biochemical methods and the transmission electron microscope was utilized to observe the ultrastructure of chondrocytes.The results showed:(1) T2 toxin could restrain both the DNA synthesis and the cell proliferation of chondrocytes in the media containing different concentrations of T2 toxin (0.001 ̄0.005mg/L).The effect of T2 toxin on the chondrocyte damage was increased with the concentration of T2 toxin in the given does.When Se was added into the media,the chondrocyte damage by T2 toxin was decreased,but can not prevented.(2)T2 toxin led to few effects on chondrocytes as its concentration was 0.0005mg/L.It caused evident damage to chondrocytes when its concentration was 0.001mg/L and 0.005mg/L and the cell damage was significantly different from that in the control group (P<0.001).
