摘要
本实验应用超数排卵,用乙烯雌酚(DES)延长发情期,并于发情后不同时间交配,光镜和电镜等方法研究了拖延交配对黑龙江地方山羊受精的影响。实验结果:(1)于排卵前6小时开始间隔12小时注射3(1+2)mg或5(2+2+1)mgDES可分别使山羊发情持续时间延长14.4和24.8小时;(2)注射乙烯雌酚对山羊排卵时间、获卵率、卵子形态及受精卵的发育都没有明显影响;(3)发情30和36小时交配山羊的受精率和胚胎的发育进程与发情12小时交配山羊受精率和发育进程没有明显差异;(4)发情42小时交配山羊受精率明显下降,3只山羊中只有1只产出2枚受精卵;发情48小时交配山羊无一产出受精卵;(5)电镜和光镜下检查受精时间后48小时未卵裂卵子未见有受精迹象,也未见有精子附着在透明带上。实验结果(1)和(2)说明:应用DES可达到延长动物发情持续时间的目的;可考虑用DES这种人工合成品取代价格贵的雌二醇—17β来进行动物发悄同步和改善超排效果等方面的研究。实验结果3说明:山羊精子可在交配后6小时左右运行到输卵管受精部位并完成获能;山羊精子和绵羊精子一样也不是在输卵管峡部贮存期间获能的;于发情后期输精的精子无需在输卵管峡部贮存便可上行获能,表明于发情末期对山羊进行配种或输精是可行的,便于人们在某些情况下小范围地调整输精时间。
To study the effcts of delayed mating on fertilization in goats,superovulated does were treated with DES to prolong estrous durationand were mated at different hours after the onset of estrus. Eggs andembryos were recovered and examined by light and electron microscopy. The results are as follows:1. A proper amount of DES given to thesuperovulated does around the time of ovulation prolonged their estrousduration and the fertilization and early embryonic developm ent were notaffected. This imply that DES can be used to replace E-17β tosynchronize estrus or improve response to superstimulation. 2.Thefertilization rate and embryo development were not affected when matingwas delayed until the time of or 6 hrs after ovulation. This indicatedt hat :(l)the goat sperm were transported the fertilization site andfinished capacitation with 6 hrs after mating aroud the time ofovulation;(2)the sperm deposited in the vagina toward the end ofestrus went directly to the ampulla without being stored in the isthmus;(3)mating was successful in the late estrus of the goat. 3. Only 1% ofthe eggs were fertilized when mating was delayed until 12 hrs afterovulation, due,most probably,to the failure of sperm transport orcapaci tation in the female tract,not to the egg aging since nospermatozoa were found either on or within the zona of the unfertilizedeggs and the egg showed no morphological changes at this hour.
