摘要
目的探讨建立人类胃癌裸鼠原位移植转移模型简便易行的方法,比较三种方法的成瘤率。方法用人类胃癌细胞系 SGC-7901悬液接种于裸鼠皮下,形成稳定传代的皮下移植瘤,连续传八代。15只裸鼠随机分为三组,以第八代裸鼠皮下移植瘤组织为移植材料,分别用捆扎法、医用 OB 胶法以及两种联合的方法把皮下肿瘤组织移植至裸鼠胃浆膜下形成原位移植肿瘤。结果 6~7周后,处死全部裸鼠,捆扎组、医用 OB 胶组和联合组成瘤率分别为4/5、3/5和5/5,捆扎联合医用 OB 胶组成瘤率最高。结论应用缝线捆扎联合 OB 胶建立人类胃癌裸鼠原位移植模型是一种简便易行的方法,值得推广和应用。
Objective To establish a simple convenient model of human gastric cancer in nude mice via orthotopic transplantation.Tumor-take rates of three different approaches were compared and analyzed. Methods SGC7901 cell suspension was subcutaneously implanted into the nude mice to develop subcuta- neous implant-tumor,the tissue of which was subcutaneously implanted into the nude mice of next generation repeatedly.Fifteen nude mice were randomly divided into the same three groups,the tissue of subcutaneous implant-tumor of eight generation was orthotopically implanted into the nude mice stomach wall in tie-up,OB biological glue and the united respectively.Results 6~7 weeks later,all of the nude mice were killed,tu- mor-take rates was 4/5 in tie-up,3/5 in OB biological glue,5/5 in the united.Tumor-take rates of the united was the highest.Conclusion The united is a simple convenient method of constructing the orthotopieally implanted model of human gastric caner in nude mice.
