摘要
介绍了一种分离纯化冈村枝管藻的方法:用组织匀浆机将孢子体磨碎,用灭菌海水稀释,在倒置显微镜下用自制的显微操作针挑出端部多室孢子囊饱满的同化丝,在盛有Provasoli培养基的试管中培养。培养温度为23℃,光周期为12L∶12D。15 d左右后,部分试管中发育出孢子体。
A kind of separation and purification method of Cladosiphon okamurarnus was introduced. The sporophytes were grinded and diluted with sterilized seawater. Under inverted microscope, well-developed plurilocular sporangia were selected with homemade microneedle and cultured in test-tube with Provasoli culture medium at 23℃ with the photoperiod of 12L: 12D.About 15 days later, sporophytes were seen in some test tubes.
