摘要
目的从富含多酚和多糖的泰山白花丹参根中分离出高质量RNA。方法比较了5种RNA提取方法,获得了一种以CTAB法为基础的分离高质量完整RNA的简便、快速方法。结果使用该方法从泰山白花丹参根部分离的RNA的纯度是:A260/A280为1.87,A260/A230为2.11,产量为284.62μg/g。FW的根,电泳条带清晰,RNA完整性好。由此法所得的RNA可直接用于cDNA文库构建等分子生物学实验。结论该技术可有效地从富含多酚和多糖的泰山白花丹参根中分离出高质量RNA,为对该山东特有药材进行分子生物学研究奠定基础。
Objective To explore the high quality total RNA isolation method from root of Taishan Salvia miltiorrhiza Bge. f. alba C. Y. WU et H.W. Li containing much polyphenols and amylose. Methods Simple and rapid procedure for high - quality total RNA isolation modified from CTAB based method was obtained by comparing five RNA isolation methods. Results A260/A280 and A260/A230 of prepared RNA were respectively 1.87 and 2.11, and RNA amount were 284.62μg/g fresh weight of root. The results of electrophoresis showed that RNA bands were clear and showed no degradation. This high quality RNA could meet the demands of molecular analysis, such as cDNA library construction. Conclusion This modified technique based on CTAB method may be ef- ficient and reliable in isolating high quality RNA from Taishan Salvia miltiorrhiza Bge. f. alba C. Y. WU et H.W. Li containing much polyphenols and amylase. This study established the base for further study of this medicinal plant found only in Shandong province.
