大青枣叶中黄酮的提取及其含量的伏安法测定 被引量:3

摘要 [目的]研究大青枣叶中黄酮提取与测定的最佳方法。[方法]以新鲜的大青枣叶片为材料,用乙醇提取其中的黄酮,研究乙醇浓度、提取时间和提取温度对黄酮提取量的影响;用芦丁标准溶液绘制微分脉冲伏安曲线后,在磷酸盐缓冲液中,用微分脉冲伏安法测定提取液中的黄酮含量。[结果]当乙醇浓度为70%、提取温度为80℃、提取时间为4 h时,黄酮提取量最高;微分脉冲伏安法测定黄酮含量具有操作简单、灵敏度好、检出限高等特点。当磷酸盐溶液的pH值为4.5时,黄酮的检出限为3.0×10-7mol/L。[结论]大青枣叶中黄酮提取的最佳条件为:用70%乙醇在80℃条件下提取4 h。微分脉冲伏安法对黄酮含量的检测效果较好。 [ Objective ] The aim was to study the optimum method on the extraction and determination of flavonoids from cottony jujube leaves. [ Method] With fresh leaves of cottony jujube as the material, their flavonoids were extracted by ethanol, the effects of ethanol concn, extracting time and extracting temperature on extraction amount of flavonoids were studied. After the differential pulse vohammetry curve was drew with the rutin standard solution, the flavonoids content in extract was determined by differential pulse voltammetry in phosphate buffer. [ Result] When the ethanol was 70%, the extracting temperature was 80 ℃ and extracting time was 4 h, the extraction amount of flavonoids was the highest. The differential pulse vohammetry by which the flavonoids content was determined had the characters of simple operation, good sensitivity and high detection limit. When the pH value of phosphate solution was 4.5, the detection limit of flavonoids was 3.0 × 10 ^-7 mol/L. [ Conclusion] The optimum extracting condition for flavonoids from cottony jujube leaves was that using ethanol to extract flavonoids for 4 h at 80℃. The detection effect on fiavonoids content by differential pulse vohammetry was better.
