高效液相色谱法测定蔬菜中五氯硝基苯残留 被引量:8

Determination of pentachloronitrobenzene residue in vegetables by high-performance liquid chromatography

摘要 目的:建立快速、简便、灵敏的蔬菜中五氯硝基苯残留的高效液相色谱检测方法。方法:蔬菜样品经丙酮/正己烷(10:90,v/v)均质提取,用浓硫酸净化,在XDB C8(150 mm×4.6 mm i.d.,5μm)色谱柱上,以甲醇/乙酸-乙酸铵溶液(10 mmol/L,pH4.5,70/30,v/v)为流动相,流速为1.0 ml/min,柱温为35℃,紫外检测波长为254 nm。结果:蔬菜中五氯硝基苯的平均回收率在83.2%~94.2%范围,RSD小于5.5%,线性范围为0.1~50.0 mg/L(r=0.9992),检出限为0.02 mg/kg。结论:建立的方法用于蔬菜中五氯硝基苯残留的测定,具有操作简便、快速、准确的优点。 Objective:To establish a simple,accurate and sensitive method for the determination of pentachloronitrobenzene residue in vegetables by liquid chromatography with ultraviolet detection.Methods:After the sample was extracted with acetone/hexane(10:90,v/v) and cleaned up with H2SO4,a satisfactory separation was achieved on a Zorbax XDB C8 column(150 mm×4.6 mm i.d.,5 μm)using methanol/acetic acid-ammonium acetate(10 mmol/L,pH 4.5,70/30,v/v) as the mobile phase at a constant temperature of 35℃ and at a flow rate of 1.0 ml/min.Detection was performed on an ultraviolet detector at 254 nm.Results:The linear range was 0.1~50.0 mg/L and the limit of quantification was 0.02 mg/kg.The extraction recoveries were 83.2%~94.2% and the RSD less than 5.5%.Conclusion:This method is simple,rapid and accurate for the determination of pentachloronitrobenzene in vegetables.
