摘要
采用酸法对黑木耳粗粉和超微粉多糖进行分离提取。研究了超微粉粗多糖DEAE-Cellul ose52离子交换层析的分离纯化工艺。样品分出两个峰,将超微粉与两个组分收集后浓缩,采用HPLC法分析多糖组成及相对分子质量分布,并做黑木耳超微粉多糖与粗粉多糖的降血脂功能动物试验。结果表明,最佳分离条件为:洗脱液浓度0.25mol/L,流速200s/mL,上样质量浓度3.5mg/mL。黑木耳经超微粉碎后多糖相对分子质量降低明显;相对于未经超微粉碎获得的粗多糖,超微粉碎后得到的多糖具有更明显的降血脂作用。
The polysaccharides of ultra-fine powder from Auricularia auriculars was extracted from coarse powder and ultra-fine powder by acid method, and the optimal purificative conditions of DEAE-Cellulose 52 Ion-Exchange Chro- matography were studied in this paper. Concentration of NaCl is 0.25 mol/L, flow rate is 200 s/mL, the concentration of the adding sample is 3.5 mg/mL. The ultra-fine powder and two components were collected and concentrated, then the constitutes of polysaccharide and the distribution of relative molecular weight were analyzied together with the ultra-fine powder by HPLC. After albinic mice were fed with polysaccharides from ultra-fine grinding powder and coarse powder of Auricularia auricular, the changes of serum triglyceride (TG), cholesterol (TC) and high-density lipoprotein cholesterol (HDL-C) in mice showed thatuhra-fine grinding powder of Auricularia auricular had stronger function of reducing blood lipid.
