摘要
在探索TTC-脱氢酶法测定大肠杆菌脱氢酶活性的最佳条件基础上,对真空、-20℃低温对大肠杆菌脱氢酶的影响进行研究。TTC法显色最佳条件为:TTC的浓度0.4%,pH值7~8,反应时间30min,测定波长485nm。真空处理5min大肠杆菌脱氢酶活性显著降低,而-20℃低温处理1h时脱氢酶活性才明显下降。因此,-20℃低温、真空处理都会导致细菌呼吸活性的下降,细菌刚放入真空的瞬间对其呼吸活性影响较大,而-20℃低温处理1h后才引起细菌呼吸活性显著下降。
In order to investigate the effects of low temperature and vacuum on respiratory rate of bacteria with TTC-dehydrogenase activity test, the dehydrogenase activity of E. coli at - 20℃ and vacuum on respiratory rate of E. coli were measured after the conditions of TTC test were explored. As resuhs,the optimal test conditions are 0.4% TTC, pH 7-8, testing time 30 min, testing wave length 485 nm. The dehydrogenase activity distinctly falls by putting E. coli cells in vacuum for 5 rain and in -20℃ for 1 h. The research concluded that the low temperature and vacuum can reduce the respiration rate of bacteria,which rudced immediately, about 5 min, in vacuum and slowly in -20℃, over 1 h.
