摘要
从人工饲养的健康仔鸡的盲肠和大肠中分离出1株对大肠杆菌有较强拮抗作用的芽孢杆菌菌株Y-1,对该菌株进行了菌落形态、培养特征观察和生理生化特性鉴定及16SrDNA序列的分析。以16SrDNA序列为基础构建了包括11株相关种属细菌在内的系统发育树,其中与11个芽孢杆菌属的菌株的16SrDNA序列的同源性为94.63%~99.35%,Y-1被鉴定为侧孢短芽孢杆菌。模拟体内高胆盐环境(胆盐含量0.3%),监测菌株的耐受能力,然后测定在人工胃液、肠液环境中作用不同时间后的存活菌数。试验结果表明,Y-1在胆酸盐、人工胃液、人工肠液中表现出了很强的耐受能力。
A strain of Bacillus Y-1 with stronger antagonistic effect on E. coli was separated from the cecum and large intestine of healthy bred chieken. The observation of colony morphology and the cultural characteristies, the identifcation of physiological and biochemical characteristics and 16 rDNA sequences analysis were nrade ou this strain. Based on 16S rDNA sequences, a phylogenetic tree with 11 strains of related bacteria was eonstrueted. The homology of Y-1 with 16S rDNA sequences of 11 strains of Bacillus spp.was 94.63 % - 99.35 % and Y-1 was identified as Brevibacillus laterosportas .The environment of high bile salt in vivo (with bile salt content of 0.3 % ) was stimulated to monitor the tolerance of Y-1. The survival ntnnber of bacteria after being inoculated in artificial gastric juice and intestinal environment for different time was determined. The test results showed that Y- 1 showed stronger tolerance in bile salt, artificial gastric juice and intestinal liquid.
