摘要
目的观察腺病毒介导神经生长因子(Ad-NGF)基因治疗对大鼠坐骨神经损伤修复的疗效。方法取SD大鼠16只,切断右侧坐骨神经并缝合,随机分为Ad-NGF治疗组、神经生长因子(NGF)局部注射组、空腺病毒载体组和生理盐水(NS)局部注射组。术后第31天行坐骨神经功能指数及神经电生理测定,Western blot检测神经生长因子蛋白质表达水平。结果腺病毒介导神经生长因子较其余3组可显著增加局部NGF蛋白质表达水平,改善坐骨神经功能指数和神经传导速度。结论腺病毒介导神经生长因子基因治疗可有效促进大鼠坐骨神经损伤后修复。
Objective To investigate the effect of adenovirus expressing nerve growth factor (Ad-NGF) on sciatic nerve injury in rats. Methods Sciatic nerve injury model was established by transection of right sciatic nerve in 16 male SD rats, and then randomly divided into 4 groups: Ad-NGF group, nerve growth factor (NGF) group, adenovirus vector group and normal sodium group. The effect of various treatments on injured sciatic nerve was evaluated by examining sciatic nerve function index (SFI) and protein expression of NGF by Western blotting, performing electrophysiological test on day 31 after operation. Results Ad-NGF increased the protein expression of NGF and improved SFI and nerve conduction velocity significantly, as compared with the other 3 groups. Conclusion Ad-NGF can effectively promote the repair of sciatic nerve injury in rats.
