摘要
                
                    目的建立特发性高草酸尿(IH)的大鼠模型。方法将第1代72只SD大鼠(雌雄各半)分别置于大鼠代谢笼中,在适应饮食5d后收集连续2d的24h尿,用离子色谱仪测定24h尿草酸排泄量。将第1代大鼠中尿草酸排泄量最高的3只雄鼠与6只雌鼠进行交配,按同样的方法检测其子代(作为近交系)的24h尿草酸排泄量,再选择尿草酸排泄量最高的3只雄鼠与6只雌鼠进行交配。依此类推,连续对每代大鼠进行检测、筛选和近交传代至第5代。结果根据第1代大鼠的检测值确定大鼠24h尿草酸排泄量的正常值范围(x±2s):雄性为(4.82±2.94)mg;雌性为(5.21±3.26)mg。第5代近交系大鼠24h尿草酸排泄量(x±s):雄性为(12.54±2.46)mg(n=16);雌性为(13.51±2.63)mg(n=16)。将24h尿草酸排泄量高于正常值范围且在2.5倍以内的大鼠定义为IH大鼠,则在第5代近交系大鼠中,雌、雄性IH大鼠的出现率均超过90%。结论该实验建立的IH大鼠模型可稳定传代,可用于IH的病因学研究。
                
                Objective To establish a model of idiopathic hyperoxaluria (IH) in rats. Methods Seventy-two Sprague-Dawley rats (36 male and 36 female) as the first generation were placed in individual metabolic cage, respectively. Two successive 24 h urine was collected to measure the 24 h urine oxalate excretion by ion chromotagraphy after the animals had been equilibrated on the diet for 5 days. The 3 male and 6 female rats with the greatest oxalate excretion in the first generation were used to breed the next generation. A similar protocol was used to select the 3 hyperoxaluria males and 6 hyperoxaluria females for inbreeding of subsequent generations. The rest may be deduced by analogy, each generation rats were measured, screened and inbreeded of subsequent generations. Results The ranges of normal values (x±2s) of 24 h urine oxalate excretion were (4.82± 2.94) mg/24 h (male) and (5.21±3.26) mg/24 h (female) respectively, established in the normal first generation rats. Urine oxalate excretion (x±s) in the fifth generation inbred rats was (12.54±2.46) mg/24 h (male, n=16) and (13.51±2.63) mg/24 h (female, n=16). IH rats were defined as 24 h urine oxalate excretion greater than the ranges of normal values, besides lower than 2.5 times of the ranges of normal values. So the incidence of IH (female and male) rats in the fifth generation inbred rats exceeded 90%. Conclusion The IH model in rats established in this study can be kept on passage and used to study the pathogenesis of IH.
