摘要
目的:建立以反相高效液相色谱法测定熊去氧胆酸胶囊中熊去氧胆酸含量的方法。方法:色谱柱为Merck Lichro-spher100 C18(125mm×4.0mm,5μm),流动相为等体积的乙腈-0.002mol.L-1磷酸二氢钠溶液(用氢氧化钠调pH2.0),流速为1.0mL·min-1,检测波长为205nm。结果:熊去氧胆酸检测浓度在0.25～1.00mg·mL-1范围内与峰面积积分值呈良好线性关系(r=0.999 7);平均回收率为99.5%,RSD=0.9%(n=9)。结论:本方法简便、准确、重现性好,可用于该制剂的质量控制。
OBJECTIVE： To determine the content of Ursodeoxycholic acid in Ursodeoxycholic acid capsules by RP- HP- LC.METHODS： HPLC was performed on Merck Lichrospher100 C18 column（ 125 mm× 4.0 mm, 5μm） .The mobile phase was acetonitrile- 0.002 mol · L^ -7 sodium dihydrogen phosphate solution （50 ： 50） （adjust pH2.0 with NaOH） at a flow rate of 1.0 mL ·min -1. The detective wavelength was set at 205 nm. RESULTS： The calibration curve of Ursodeoxycholic acid was linear within the range of 0.25-1.00mg· mL ^-1（r =0.9997） .The average recovery was 99.5% and RSD was0.9% （n =9） CONCLUSION ： This method is simple, feasible and reproducible, and it can be used for the quality control of Ursodeoxycholic acid capsules.
