摘要 目的:探讨308nm准分子激光对T淋巴细胞直接和间接诱导凋亡的作用及转化生长因子β1(TGF-β1)在诱导T淋巴细胞凋亡中的作用。方法:308nm准分子激光分别照射角质形成细胞和体外分离的活动期白癜风患者外周血T淋巴细胞;ELISA检测照射后24h及48h角质形成细胞培养上清中的TGF-β1浓度;流式细胞仪检测照射后48h的T淋巴细胞凋亡情况;建立Transwell细胞共培养系统(角质形成细胞位于上室,T淋巴细胞位于下室),308nm准分子激光照射Transwell上室,照射后48h检测T淋巴细胞凋亡情况;照射前加入不同浓度的抗TGF-β1抗体(25、250、2500pg/L),于照射后48h采用流式细胞仪分别检测T淋巴细胞凋亡情况。结果:308nm准分子激光诱导角质形成细胞分泌TGF-β1的增加呈剂量依赖性;流式细胞仪检测结果显示:准分子激光可直接诱导T淋巴细胞凋亡;准分子激光照射Transwell上室可间接诱导T淋巴细胞凋亡;照射前加入抗TGF-β1抗体,可抑制308nm准分子激光的诱导凋亡作用。结论:308nm准分子激光直接照射可诱导T淋巴细胞凋亡,也可作用于角质形成细胞间接诱导T淋巴细胞凋亡。TGF-β1在308nm准分子激光诱导T淋巴细胞凋亡中起重要作用。 Objectives: To investigate the direct and indirect apoptotic pathways of T cells triggered by 308 nm excimer laser, and the role of TGF-β1 in inducing the apoptosis of T lymphocytes. Methods: Keratinocytes and lymphocytes separated from patients with active vitiligo were irradiated by 308 nm excimer laser respectively. Twenty-four and 48 hours after irradiation the levels of TGF-β1 in the supernatant in the medium for culturing keratinocytes were evaluated by ELISA, and apoptosis of T lymphocytes was evaluated by flow cytometry 98 hours after irradiation. Transwell cell cocuhure system was established in which keratinocytes were located on the upper chamber, and T lymphocytes in the lower chamber. The upper chamber of transwell was irradiated by 308 nm excimer laser, the apoptosis of T lymphocytes was evaluated 48 h after irradiation. Different levels of anti-TGF-β1 antibody (25, 250, 2 500pg/L) were added to the transwell system, apoptosis of T lymphocytes were evalu-ted 48 h after irradiation respectively. Results: The increase of keratinocyte-secreted TGF-β1 induced by 308 nm excimer laser was in a dose-dependent manner. Shown by flow cytometry, T lymphocyte apoptosis was found not only directly by the irradiation of excimer laser, but also indirectly by the irradiation of excimer laser to the upper chamber of transwell system. The adding of anti-TGF-β1 antibody could suppress the apoptosis of T lymphocytes induced by excimer laser. Conclusion: 308 nm excimer laser directly and indirectly induces the apoptosis of T lymphocytes, in which TGF-β1 plays an important role.
