摘要
目的比较流感病毒对Vero细胞和MDCK细胞敏感性的差异及影响敏感性的因素,寻找流感病毒灭活试验的最佳细胞。方法用测定两种细胞感染病毒后TCID50值和血凝效价值的方法,了解流感病毒对两种细胞敏感性的差异。结果Vero细胞和MDCK细胞接种流感病毒72 h,测得血凝效价分别为1:64和1:128;MDCK细胞的病变较Vero细胞明显。终浓度为5μg/ml的胰酶可提高两种细胞增殖流感病毒的血凝效价,但对TCID50值的大小无明显的影响。随着维持液中血清浓度的升高,两种细胞增殖病毒的血凝效价逐渐降低,MDCK细胞染毒后的TCID50值也明显减小。结论流感病毒对MDCK细胞的敏感性高于对Vero细胞的敏感性,胰酶和血清能影响病毒对细胞的敏感性。
Objective To compare the sensitivity of Vero cell and MDCK cell to influenza virus and the factors in fluencing the sensitivity and to find the suitable cell for use in influenza virus inactivation test. Methods TCID50 test and hemagglutinatian test were used for comparing the sensitivity of the two kinds of cells to influenza virus. Results At 72 h after inoculation of influenza virus, the titers of hemagglutination in Vera cells and MDCK cells were 1:64 and 1:128 respectively. The cytopathic effect on MDCK cells was more obvious than on Vero cells. Pancreatin at the concentration of 5μg/ml increased the titers of hemagglutination of the two kinds of cells to influenza virus, but had no effect on TCID50. Increase in sera in the maintenance media decreased the titers of hemagglutination of the two kinds of cells to influenza virus, and the value of TCIDs0 of MDCK cells to influenza viruses. Conclusion The sensitivity of MDCK cell was higher than that of Vero cell. Furthermore, pancreatin and sera could influence the sensitivity of two kinds of cells to influenza virus.
