摘要
本文取1.0-5.0mm的花蕾,用1%龙胆紫染液压片法制片,在光学显微镜下观察花粉发育情况。通过对油菜花粉细胞发育过程的观察,探讨花蕾长度与花粉发育时期的相互关系,以便在进行有关遗传性状改良或其他生物技术研究活动时,可以直接依据花蕾长度,判断花粉发育时期。结果表明:当花蕾1.0-1.5mm长时,花粉基本处于减数分裂时期;花蕾1.6-2.2m长时,为花粉发育的四分体时期;花蕾2.3-3.0mm长时,为花粉发育的单核高峰期;花蕾3.1-3.7mm长时,为花粉发育的双核高峰期;花蕾3.8-5.0mm及以上长时,为花粉发育的成熟期。
Staining different pollen grains from flower buds 1.0 -5.0mm long with 1% gentian violet and by estimating their development stages under optical microscope, the relationship between length of bud and pollen development stages in rape was studied by observing flower buds of rape of cell morphology at different pollen development stages, so we can use it in carrying on the related hereditary feature improvement and other biological technology research , we may judge pollen development stages by the length of bud. The results showed that most of pollens from the buds 1.0 - 1.5 mm long were in the stage of meiosis, and 1.6 -2. 2 mm long were in the stage of tetrad, while the peak stage of single - nucleus, double - nucleus and mature period pollens appeared when the buds were 2. 3 -3. 0mm,3. 1 -3. 7mm and 3. 8 -5. 0mm long, respectively.
