促进干细胞增殖的龟版浸膏气相色谱-质谱指纹图谱分析与应用 被引量:1

摘要 目的运用生物活性指纹图谱预测其它中药能否促进骨髓间充质干细胞(bone marrow mesenchymal stem cells,bMMSCs)。方法龟版浸膏依次用石油醚、乙醚、二氯甲烷提取,得到溶剂提取物,用MTT比色法及流式细胞仪器研究了溶剂提取物干细胞活性,采用GC-MS技术研究每一个提取物,根据总离子流色谱图的相同保留时间和相应峰面积,获得龟版浸膏GC-MS生物活性指纹图谱。结果初步结果表明溶剂提取物均能促进鼠bMMSCs的体外增殖,用龟版浸膏GC-MS生物活性指纹图谱来预测中药"四物汤"的干细胞活性,预测准确率达83.3%。结论用这种方法来预测未知中药样品的干细胞活性,有一定的可靠性。 Objective To predict the promoting effect of Chinese medicine on proliferation of rat bone marrow mesenchymal stem cells(bMMSCs) by bioactivity fingerprint in vitro. Methods The ointment of plastrum testudinis was extracted by petroleum ether, ether and dichloromethane in bum to gain the extracts. The effects on the proliferation of bMMSCs were examined by MTT assay and flow cytometry analysis, and each sample was studied by GC - MS. According to the total ion current chromatogram of the same retention time and the corresponding peak area, the GC - MS fingerprint of bioactivity was gained. Results The results showed that the substances extracted could promote the proliferation of stem cells. The fingerprint of bioactivity was used to evaluate the bioactivity of "Si Wu Tang", and the accuracy rate reached 83.3%. Conclusion This method is reliable to predict unknown Chinese medicine samples of stem cell activity.
