摘要
目的建立适合人二倍体细胞培养血清的筛选方法。方法通过测定细胞倍增时间和进行细胞生长促进试验对15批次的新生牛血清作了筛选,并将筛选出的11批次血清用于大规模细胞培养,观察细胞的生长维持情况。结果发现细胞倍增时间与相对生长率无明显相关性(P>0.05)。在大规模细胞培养中,相对生长率97%以上的血清更适合二倍体细胞生长。结论细胞生长促进试验比细胞倍增时间测定更能客观地反映血清促人二倍体细胞的生长能力。细胞生长促进试验可能是较佳的。
Objective To establish a screening method of serum suitable for culture of human diploid cells. Methods Two methods of determination of cell doubling time and cell growth promotion assay were used to screen the newborn calf serum (NCS). A total of 11 in 15 different hatches of NCS were selected and used for large-scale cell culture so as to observe the cell growth status. Results There was no significant correlation between the cell doubling time and relative growth rate ( P 〉 0.05). The NCS which the relative growth rate of cells cultured in was above 75% ,was mere suitable for the growth of human diploid cells in large-scale culture. Conclusion The method of Cell growth promotion assay is better than the one of determination of cell doubling time in reflecting the growth ability of human diploid cells in the serum. Cell growth promotion assay is a mere suitable method for screening NCS used for human diploid cells culture.
