摘要
为研究鸡白细胞介素2对新城疫La Sota弱毒苗免疫效果的促进作用,纯化真核重组质粒pcDNAChIL-2,细胞转染验证后,将重组质粒分为0,5,10,20和30μg5个剂量组,分别与LaSota弱毒苗同时免疫7日龄雏鸡,免疫后7d,每隔1周分离鸡血清,测量血清中NDV的HI效价。结果表明,混合注射有pcDNAChIL-2重组质粒雏鸡的NDV抗体产生的水平显著升高,其中以20和30μg剂量组最为明显,提示ChIL-2可有效促进La Sota弱毒苗的免疫效果。
In order to investigate the immunization effect of attenauted La Sota vaccine facilitated by ChIL-2,the ukaryotic recombinant pcDNAChIL-2 was purified and the expression was validated by cell transfection in vitro. Five groups with one dose of 0,5,10,20 and 30 μg plasmid was designed,which immunized with one dose of La Sota vaccine together. 7 days post immunization,the serum was collected every week to detect the antibody against the NDV by hemagglutinin inhibition test. The results showed that the pcDNAChIL-2 could boost the antibody level effectively,and the group with 20 μg and 30μg plasmid were most effective,which indicate the ChIL-2 could improve the antibody level of La Sota vaccine effectively.
