杜仲提取物抗HIV活性成分的分离鉴定 被引量:8

摘要 目的:研究杜仲提取有效成分对HIV gp41六股螺旋束的抑制作用,为寻求新的抗HIV病毒进入抑制剂提供实验依据。方法:将杜仲水提取液分别用石油醚、乙酸乙酯和正丁醇萃取,萃取物干燥称量后制备成浓度为1mg/ml的样品液,用夹心免疫酶联法检测杜仲及其萃取物对HIV gp41六股螺旋束结构形成的抑制作用。结果:发现杜仲抗HIV gp41的活性部分存在于在乙酸乙酯萃取物中,经过醇沉后的上清液失去活性,而沉淀物具有较高的活性。结论:杜仲可抑制HIV gp41六股螺旋束的形成从而起到抗HIV的作用,其活性部位主要在乙酸乙酯萃取物里,活性物质是能被乙醇沉淀的大分子物质。 Objective : To investigate the anti-HIV effects of Eueommia ulmoides Oliver, so as to provide experimental basis for searching a new efficacious drug for treatment of AIDS. Methods: Using phytoehemistry to isolate compounds from Eucommia ulmoides Oliver, the inhibitory activity of Samples on the HIV gp41 six-helix bundle formation was determined by a modified sandwich ELISA and PAGE. Results : The Samples from Eucommia ulmoides Oliver had potent inhibitory activity on the HIV gp41 six-helix bundle for- mation. Conclusion: Eueommia uloides Oliver can inhibit HIV by targeting HIV gp41.
