摘要
目的探讨基质金属蛋白酶MMP-2和MMP-9与多发性肌炎,皮肌炎(PM/DM)的关系。方法采用实时荧光定量PCR和Western印迹方法分别检测PM/DM患者外周血单一核细胞(PMBC)中MMP-2和MMP-9的mRNA和蛋白表达水平。结果MMP-2、MMP-9在PM/DM患者外周血PMBC中的mRNA表达水平明显高于正常人对照组,其中MMP-2为正常人对照组的2.41倍、MMP-9则为1.66倍。Western印迹检测发现MMP-2、MMP-9在PM,DM患者外周血PMBC中的蛋白表达水平也明显高于正常人对照组,与其mRNA表达结果一致。结论基质金属蛋白酶MMP-2和MMP-9可能在PM/DM的发生中起作用。
Objective To investigate the relation of the expressions of matrix metalloproteinase-2 (MMP-2) and -9 (MMP-9) with polymyositis/dermatomyositis (PM/DM). Methods Blood samples were obtained from 8 patients with PM, 12 patients with DM and l0 normal human controls. Real-time quantitative RT-PCR and Western-blot were applied to examine the mRNA and protein expressions of MMP-2 and MMP-9 respectively in the peripheral blood mononuclear cells (PBMC) of these patients. Results The mRNA expression levels of MMP-2 and MMP-9 in the PBMC of patients with PM or DM were 2.41 and 1.66 folds as high as those of the normal controls, respectively; these differences were significant between the two groups. Increased expressions of MMP-2 and MMP-9 were also noted in the patients with PM or DM. Conclusion MMP-2 and MMP-9 may play a part in the development of polymyositis and dermatomyositis.
