摘要
目的观察秦艽水煎液(Gentiana water exact)对家兔全脑缺血再灌注损伤模型热休克蛋白70(HSP70)表达的影响,探讨Gentiana water exact对全脑缺血再灌注损伤的保护机制。方法参照Pulsinelli等造模方法进行改良,结扎双侧椎动脉,夹闭双侧颈总动脉,制成"四管闭塞法"全脑缺血模型。缺血5 min后分别于再灌注12 h、24 h、48 h取材,使用免疫组化技术检测家兔全脑缺血再灌注损伤模型双侧海马CA1区HSP70的表达。结果Sham组无表达;I/R组与G组阳性表达,且随灌注时间的延长而表达增强,24 h达到高峰;与I/R组相比,G组各时间点的HSP70表达明显增多(P<0.05)。结论秦艽可以通过对HSP70表达的上调,达到对脑损伤的保护作用。
Objective To investigate the effects of Gentiana water extract on the heat shock protein 70 expression in global cerebral ischemia-reperfusion injury in rabbits.Methods According to the Pulsinelli model building method,we legate bilateral vertebral artery and clamp bilateral CCA,use four-vessel occlusion method to make the complete cerebral ischemia-reperfusion model.After global cerebral ischemia established for 5 minutes,we bring out the organization of different group at reperfusion 12h,24h,48h respectively and use immunohistochemistry to detect the HSP70 expression in bilateral hippocampus CA1 section.Results There is no expression of HSP70 in the Sham group,the expression in the G is significantly higher compared with the I/R at above point(P〈0.05),there is a increasing trend on HSP70 expression with reperfusion time extension,the expression reached summit at ischemia-reperfusion 24h,then begins to decrease. Conclusion Gentiana water extract can evidently increase HSP70 expression in global cerebral ischemia-reperfusion injury in rabbits as a consequence to protect nerve cells in cerebral ischemia-reperfusion injury.
