木瓜中等极性部位高效液相色谱指纹图谱研究 被引量:4

摘要 目的研究不同采收时间和不同炮制方法的木瓜药材中等极性部位指纹图谱,为木瓜的规范化种植与合理应用提供依据。方法以咖啡酸、绿原酸为参照物,采用高效液相色谱(HPLC)法测定分析各指纹图谱,色谱条件:C18(Kromasil C18,10nm^5μm,4.6mm×250mm)色谱柱;流动相:(A)乙腈-甲醇(6:4),(B)0.05%磷酸梯度洗脱,0~12min(10%~30%A,90%~70%B),~30min(30%~50%A,70%~50%B),~35min(50%~70%A,50%~30%B),~45min(70%~90%A,30%~10%B),~50min(90%~60%A,10%~40%B),~60min(60%~10%A,40%~90%B),~70min(10%A,90%B);流速:0.8mL·min-1;检测波长:283nm。结果木瓜中等极性部位指纹图谱有较好的相关性,木瓜不同采收时间的样品分为3大类,其中1~4批为一类,具有2个共有峰,5~7批为一类,具有3个共有峰,第8,9为一类,有3个共有峰;其中又以第5批采收时间最好,其次是第6,7批;不同炮制方法以晒制方法特征峰峰面积大;存放时间对于成分含量也有一定影响。结论该研究有助于木瓜的质量控制、采收和应用。 Objective To study the fingerprint of middle polarity part from Fruit of Chaenomeles at different time and in different process methods, which could provide GAP basis for planting and reasonable application. Methods HPLC with UV detector was used to analyze the patterns of fruit of Chaenomeles. Coffeic acid and chlorogenic acid were used as standard substances, Cls (Kromasil C18 10 nm- 5 μm,4.6 mm × 250 mm)column was used with a mobile phase of (A) acetonitrilemethonal (6:4)(B) 0. 05 % phosphoric acid gradient elution , 0-12 min (10 % -30%A, 90% -70 %B) , -30 min (30% -50 %A ,70% -50%B) , -35 min (50% -70 %A,50 % -30%B), -45 min (70% -90%A ,30% - 10%B) , -50min (90%-60 %A ,10%-40%B) ,-60 min(60%-10%A ,40%-90%B) , -70 min(10%A ,90% B) ;The flow rate was 0.8 mL · min^-1. The wavelength of detector was set at 283 nm. Results The fingerprint of fruit of Chaenomeles was showed an excellent correlation. Samples of Chaenomeles fruit collected from different time were divided into 3 types, 1 - 4 shared 2 common peaks, 5 -7 shared 3 common peaks,8 and 9 had 3 common peaks. Collecting time of the 5th bath is the best time for the 5th batch;the typical peak volume was the largest for the process of drying in the sun. Conclusion The study is helpful for the quality control, the gathering and application of Chinese flowering
